I hold that Calvin and Hobbes was the best comic strip.
Far Side is the best comic overall, but it was not a strip.

Liberty Meadows and Pearls before Swine also have their moments.

And if you're bored on the internet check out www.xkcd.com, he's good for a grin if you're in on the joke.

Calvin and Hobbes, definitely . Currently, I like Non Sequitor and Rose is Rose, as well as Pearls before Swine.

Most definitely, hands down, the best comic strip ever was Pogo. 

My second favorite was Li'l Abner.

Probably couldn't get away with that stuff today.  Not very PC, yanno.

I've got to cast a vote for Get Fuzzy.....probably because I live with his not-quite-as-evil long lost second cousin!

Rose is Rose is a good one too....but I don't read the comics much, so I'm not sure what else is out there.

Calvin and Hobbes is a great comic.
