I do this for a living, make ready and painting apartments between tenants. I never use tape or tarps, becasue I have found them to make more of a mess. I keep my paint tray on a plastic bag on the kitchen counter to minimize bending and practice mindfullness with the rollers and brushes. I do the whole apartment that way. Its pretty easy to paint straight lines, but you might want to cover or tape around any fancy light fixtures or expensive woodwork, especially if its in a tight corner or akward space. You can get small rollers with narrow trays for them, and those come in really handy for small rooms, spot painting, and tight spaces, behind the toilet narrow spaces between cabinets ect. You get the manuverability of a brush with the speed of a roller, I have been known to paint a whole room with just these. If you do the ceiling, do that first and keep in mind that if you have a rough textured ceiling, it will cause the roller to emmit a fine spray that will cover everything in the room including you! Also take it in stages and take your time, you can really strain your neck and shoulders doing this. Stop and strech often. (I HATE painting ceilings!) Usualy I am painting right over the same color, so I dont need primer. I have used paint with primer in it, every now and then a tenant paints a wall or two some other color. But found I just have to paint again over it with our standard white to make it match the rest of the unit, so now I just use Kilz when that happens, its cheaper. Kilz also kills mold, so if you have problems with that I would definantely use it before painting. I have a lot of fun doing this, sometimes Shepard gives me nasty little "assignments" do do while at work. Makes the work day a little more interesting I say.
