DNA testing is done by extracting a sample and using DNA polymerase to create more of the molecules. Step 2. Use a 'restriction enzyme' that recognizes certain sequences and cuts the DNA at those sequences. As everyone's DNA is different, the cuts will yield parts of different lengths. Step 3. Place the mixture of DNA fragments on a semipermeable gel and place a small DC charge across the gel. Step 4. The fragments will migrate thru the gel in reverse proportion to their Charge/Mass ratio, (the DNA has a lot of phosphate in it which has a negative charge) Preliminary results can be obtained in 12-24 hours. If you need a fast quick and dirty, you slightly increase the DC voltage which makes the fragments move a bit faster. How do I know? I did it when I was in grad school.

Yeah... Obama was killed in the last 48 hours? Let's see a show of hands who believes that. We ran Sadaam Hussein down in no time flat, Obama takes us ten years. We have less of a presence in the region than just a few years ago and yet....viola....we suddenly found him, killed him and buried him at sea. It all sounds like a bunch of bullshit. And the point that I want to make with this post is to look at how many people think it is bullshit! Thirty years ago, 90% of Americans would have believed it and the soldiers that took him out would have been given a ticker tape parade. Now we hear the news and far too many of us shrug our shoulders and say "maybe so...but maybe not." Our faith in our leaders is at an all time low and for good cause! Skepticism and/or ambivalence should be the reaction of the fringe and its not. Usama's alleged death has come when Obama needs it most; on the cusp of $5.00 per gallon gasoline, another plunge in real estate prices, a multi-billionare fucking with him to no end, an all time low in his approval ratings, etc. etc. etc. And....like 9-11....Usama's death is neat and clean and fits in a pretty little box with a lovely bow and all of our questions are supposed to be answered and all of our fears assuaged. It isn't that simple.

My thanks go out to all the unrecognized people who made it possible, from the Pakistani informant who gave up the location of the Courier, to the CIA Officer who acted on it and sent it up the chain of command, and the SEALS who completed the mission without taking a casualty, and the entire JSOC command. The President deserves credit here as well, although he'll get plenty of recognition for the take down. Good job Mr Obama.

What sort of setup, resources, environment did you need to ensure the results would not be contaminated and possibly skew the results? starshine All you need is one small sample to start with and the DNA polymerase will make you as much identical DNA as you want for multiple runs. As for resources, a well equipped university lab. As for making sure the DNA wasn't contaminated. See above. This wasn't a commercial lab where the folks may or may not know which end of a microscope to look into. We (along with other universities) were doing the preliminary research that DNA identification used by police agencies all over the world is based on. It's like any other test. You can get good results FAST if the people involved are experienced analysts and not techs. the commercial DNA testing protocols are written so that techs with moderate skill levels can obtain reproducable data that is good enough for a court case. More competent and experienced lab personnel can obtain data that is at least as, if not more, precise much faster. I have a feeling they wouldn't trust Bin Laden's (presumed) DNA to some college kid working part time.

Once you run the PCR test you get a sheet with the various markers represented. You can compare that sheet to another done at a different time and place.

His sister had brain cancer and was treated and died at Mass. General Hospital in Boston. Her DNA was taken when she died and has been on file here in Boston for a while. That's the DNA sample they matched it to. QuIte simple really. And before you pull out a conspriacy theory on that .. here's my answer: Yes, he has plenty of living brothers and sisters and family members who they could have gotten DNA from. But since they already had a DNA profile on hand, why would they waste more time and money in seeking out and gathering up more DNA. If you have it, use it. mbmbn
